RRC-CCVRPC: Xenogen IVIS Spectrum
The Xenogen IVIS® Spectrum is an advanced optical imaging system for non-invasive longitudinal monitoring of disease progression, cell trafficking and gene expression patterns in live mice, relying on quantitative detection of light emitted from both bioluminescent and fluorescent reporters. The Spectrum can be used in oncology research to follow disease progression, to detect metastasis, and to monitor responses to therapeutic treatments. Other areas of research include: infectious disease, inflammation, metabolic disease, neurology, gene therapy, stem cell biology, cardiovascular disease, immunology, and drug metabolism. Ex vivo imaging of tissues and organs is also possible, as is analysis of light emission from cells in culture. The Spectrum is made by Caliper Life Sciences, now a part of Perkin Elmer. IVIS Spectrum User Policies
The IVIS Spectrum is located in room 112 of the BRL. Researchers wishing to use the Spectrum for fluorescent or bioluminescent imaging of mice should contact Dr. Jeanette Purcell, clinical veterinarian, to confirm that their animals are permitted in the imaging room. Once an imaging study is initiated animals are not to be returned to their home room; rather, they must be held in room 122 for the duration of the study. It is the responsibility of the investigator to make the necessary arrangements for transporting animals into these rooms. In vitro imaging options are also available. All imaging studies involving live animals and/or biohazardous material require an approved ACC Animal Protocol and/or IBC Institutional Biosafety Protocol.
Every user must receive training from the staff of the Center for Cardiovascular Research Physiology Core (CCVRPC). Each 2.5- to 3.0-hour training session includes procedures for animal handling and isoflurane anesthesia; basic imaging techniques; data storage and analysis; and instrument and equipment sanitization. Each trainee will receive an imaging guide containing instructions and important contact information. Under no circumstance is an authorized user permitted to train a colleague. Violators will forfeit their access to the instrument. Once an investigator has been trained s/he will have independent use of the imager. Assisted use is also available; please note, animal preparation, including reagent injections and fur removal (if necessary), is the responsibility of the investigator.
To schedule time on the instrument each trained user must submit to the CCVRPC Director the applicable approved ACC/IBC Protocol number(s). Following confirmation, the user will be granted access to UICore, which allows time to be scheduled on the IVIS. Access to UICore requires that you be registered with the RRC. The IVIS log book, which is located at the instrument station, must be used to record the time at the BEGINNING and END of every imaging session.
Each user shall create one or more desktop folders on the instrument computer into which all data from the current imaging session may be saved. Once the session is over the folder(s) must be "zipped" and uploaded to one of three available cloud storage services (located in the Startup menu): Drop Box, Google Drive, U of I Box. Folders may also be temporarily saved in the desktop "Users Folder;" however, do not use the instrument computer as your data archive. The CCVRPC periodically will delete files and folders to free up disk space.
Data analysis should not be performed on the instrument computer. Rather, a separate workstation is available for use. Under no circumstances is the installation of additional software onto either computer allowed.

Sign-up and Cancellation Policies:
Upon successful completion of training and submission of the applicable approved ACC and/or IBC Protocol number(s) to the CCVRPC Director the user will be granted access to UICore to schedule time on the IVIS. Cancellations must be made before the reservation start-time or you will be charged. The instrument is available for use 7 days a week from 7:00 A.M. to midnight. If you require CCVRPC staff assistance during your session please make sure a CCVRPC staff member is available before you reserve your time.
There is a minimum 30-minute charge, and additional time is billed in 30-minute intervals. Users are charged for the entire scheduled time, even if they leave early. Exceptions to this policy will be considered on a case-by-case basis upon consultation with CCVRPC staff. You are welcome to stay past your scheduled time if no one else has signed-up, but you will be charged for the extra time. Regardless, every user MUST sign in and out of the IVIS log book provided at the instrument station. A $25.00 non-compliance fee will be charged for any imaging session that has not been scheduled through UICore.
Data analysis should not be performed on the IVIS instrument computer. Remote access to a separate computer workstation for image analysis is available to approved users. Unlike the IVIS computer, each user will be given an individual login (account sharing is not allowed). Remote access instructions are available online and e-mailed to all new remote access users.
The IVIS instrument computer has limited network access, which includes options for cloud data storage. You are responsible for uploading your data to one of these services (located in the Start menu) for safe-keeping and for use elsewhere.
Please acknowledge use of the IVIS Spectrum imaging facility, UIC Research Resources Center in all publications. In addition, co-authorship of papers may be appropriate when a facility staff member provides significant input to a final research outcome; if not co-authorship, a personal acknowledgment may be warranted.
Publication of data is an important way of measuring and documenting the success of the facility. If you publish any data acquired on the Spectrum please notify the director so it may be added to our publications list.
